L-Phenylalanine (Phe) is an essential amino acid that cannot be synthetized by humans or other vertebrates and must be supplied from an exogenous diet. Phe is transformed to L-Tyrosine (Tyr) through the Phenylalanine hydroxylase (PAH) enzyme and thus serves as a precursor of key catecholamines including Dopamine, Noradrenaline and Adrenaline. Mutations in the gene encoding for PAH have been linked to Phenyloketonuria (PKU), an aminoacidopathies – associated with fair skin, eczema, seizures, tremors, and hyperactivity – characterized by an increased blood/brain level of Phe and its derivatives, and a concomitant decrease in Tyr. Besides its known role in PKU, Phe has been recently linked to severity of COVID-19 where higher level of Phe has been detected for patients with moderate/severe disease in comparison to patients with asymptomatomatic/midly SARS-COV-2 infection (Atila et al, Amino Acids, 2021) thus shedding light on role of Phe on the control of immune response.
