Rinse the organism briefly in cold PBS, blot dry, then homogenise (10–20 µL ice‑cold PBS + 0.1 % ascorbic acid per mg wet weight). Centrifuge 10 min / 4 °C / 10 000 g and use 1 µL supernatant per well, diluting further if values exceed the 2.5 ng/mL top standard. The 5 pg/mL LOD and wide 0.015–2.5 ng/mL range allow substantial dilution head‑room.
