quinolinic-acid_kynurenines_1028Quinolinic acid (QA) is a downstream metabolite of the kynurenine pathway, which converts the amino acid tryptophan into nicotinamide adenine dinucleotide (NAD+), a co‑factor for numerous enzymatic reactions. QA is neurotoxic, chiefly through activation of the N‑methyl‑D‑aspartate (NMDA) receptor. Its production in myeloid‑derived cells (e.g., macrophages and microglia) is strongly up‑regulated by inflammatory stress, implicating QA in neuroinflammatory disorders such as Alzheimer’s and Parkinson’s diseases, multiple sclerosis, and amyotrophic lateral sclerosis. Proposed as a biomarker for these conditions, QA also warrants investigation as a surrogate marker in other inflammation‑driven diseases, including cancer and metabolic disorders.
